Zilbervarg I.R. The biotechnology basis of nep (Nepeta sp.) polyploid plants obtaining with application of antimicrotubule agents for the purposes of breeding. - Manuscript.
Thesis for degree of Doctor of Philosophy (PhD) by speciality 03.00.20 - biotechnology. Nikitsky Botanical Garden - National Scientific Centre, Yalta, 2002.
The work is devoted to study of biotechnological bases of nep (Nepeta sp.) polyploid plants obtaining with application of antimicrotubule agents. Optimum terms of nep explants introduction in conditions in vitro were revealed, the factors of microshoots propagation for further polyploidization and breeding were determined. High poliploidy activity of dinitroanilin and phosphorothioamid herbicids was confirmed. It was established, that dinitroanilins and APM are more effective, then colchicin and caused polyploidization in concentration on some orders below. The optimum concentration of antimicrotubule agents in liquid culture mediums for pretreatment was 60-120 мM, in agar-solidified culture mediums for treatment during of the cultivation - 10 мM.
During of the work the forms N.transcaucasica (initial 2n = 2х = 18 chromosomes), containing 2n = 4x (36), 8x (72), 16x (144), N.grandiflora - with 2n = 4x (72), 8x (144), 16x (288), and also of chimerical plant were obtained. From the hybrid form (N.cataria x N.grandiflora) x N.transcaucasica (initial number chromosomes 2n = x1 + x2 + x3 = 45) the polyploid clones c 2n = 2 (x1 + x2 + x3), 4 (x1 + x2 + x3) were obtained.
Key words: nep, in vitro, polyploid, colchicin, dinitroanilin herbicids, amiprophosmethyl.
